Paradoxical and contradictory effects of imatinib in two cell line models of hormone-refractory prostate cancer.

Imatinib mesylate is a chemotherapeutic drug that inhibits the tyrosine kinase activity of c-KIT and has been successfully used to treat leukemias and some solid tumors. However, its application for treatment of hormone-refractory prostate cancer (HRPC) has shown modest effectiveness and did not follow the outcomes in cultured cells or animal models. Moreover, the molecular pathways by which imatinib induces cytotoxicity in prostate cancer cells are poorly characterized.

Baicalein attenuates proteasome inhibition-induced apoptosis by suppressing the activation of the mitochondrial pathway and the caspase-8- and Bid-dependent pathways.

Impairment in ubiquitin-proteasome system has been shown to be implicated in the pathogenesis in neurodegenerative disorders, such as Parkinson's disease. Flavonoid baicalein has demonstrated anti-oxidant and anti-inflammatory effects. However, the effect of baicalein on the neuronal cell death due to proteasome inhibition has not been studied. Thus, in the respect of the cell death process, we assessed the effect of baicalein on the proteasome inhibition-induced apoptosis using differentiated PC12 cells. The proteasome inhibitors MG132 and MG115 induced a decrease in Bid, Bcl-2, Bcl-xL and survivin protein levels, an increase in Bax levels, loss of the mitochondrial transmembrane potential, release of cytochrome c, activation of caspases (-8, -9 and -3), an increase in the tumor suppressor p53 levels and cleavage of PARP-1. Baicalein attenuated the proteasome inhibition-induced changes in the levels of apoptosis-related proteins, formation of reactive oxygen species, depletion of GSH, DNA damage and cell death. The results show that baicalein may attenuate the proteasome inhibition-induced apoptosis in PC12 cells by suppressing the activation of the mitochondrial pathway and the caspase-8- and Bid-dependent pathways. The preventive effect appears to be attributed to its inhibitory effect on the formation of reactive oxygen species and depletion of GSH.

Some components of animal venoms have been identified that possess substantial anticancer properties. In our previous studies, the cytotoxic effects of ICD-85 (venom-derived peptides) have been reported on HL-60 and MDA-MB231 cell lines. This has prompted us to investigate the comparative cytotoxic effects of ICD-85 on the HeLa cell line and normal lamb kidney (LK) cells.

[Heat shock protein 90--modulator of TNFalpha-induced apoptosis of Jurkat tumor cells].

rTNFalpha-induced programmed death of Jurkat tumor cells cultured with 17-AAG, a selective inhibitor of heat shock protein (Hsp90), was studied by fluorescent microscopy with the use of FITC-labeled annexin V and propidium iodide. Caspase-3 and -8 activities were determined by spectrophotometry using a caspase- 3 and -8 colorimetric assay kit. It was shown that inhibition of Hsp90 leads to activation of Jurkat cell apoptosis while Hsp90 itself suppresses this process. 17-AAG enhances rTNFa-induced apoptosis of tumor cells.

Silencing of high mobility group A1 enhances gemcitabine chemosensitivity of lung adenocarcinoma cells.

The high mobility group A1 (HMGA1) proteins are architectural transcription factors found to be overexpressed in lung adenocarcinoma. Lentivirus-mediated RNA interference (RNAi) technology is a powerful tool for silencing endogenous or exogenous genes in human cancer cells. Our preliminary study shows that gemcitabine inhibits growth of the human lung cancer cell line SPCA-1 and induces apoptosis, and this effect might link with down-regulation of HMGA1 expression.
