Advances in miniature CRISPR-Cas proteins and their applications in gene editing.

The CRISPR-Cas system consists of Cas proteins and single-stranded RNAs that recruit Cas proteins and specifically target the nucleic acid. Some Cas proteins can accurately cleave the target nucleic acid under the guidance of the single-stranded RNAs. Due to its exceptionally high specificity, the CRISPR-Cas system is now widely used in various fields such as gene editing, transcription regulation, and molecular diagnosis. However, the huge size of the most frequently utilized Cas proteins (Cas9, Cas12a, and Cas13, which contain 950-1,400 amino acids) can limit their applicability, especially in eukaryotic gene editing, where larger Cas proteins are difficult to deliver into the target cells. Recently discovered miniature CRISPR-Cas proteins, consisting of only 400 to 800 amino acids, offer the possibility of overcoming this limitation. This article systematically reviews the latest research progress of several miniature CRISPR-Cas proteins (Cas12f, Cas12j, Cas12k, and Cas12m) and their practical applications in the field of gene editing.

Plant diversity increases spatial stability of aboveground productivity in alpine grasslands.

Plant diversity can significantly affect the grassland productivity and its stability. However, it remains unclear how plant diversity affects the spatial stability of natural grassland productivity, especially in alpine regions that are sensitive to climate change. We analyzed the interaction between plant (species richness and productivity, etc.) and climatic factors (precipitation, temperature, and moisture index, etc.) of alpine natural grassland on the Qinghai-Tibetan Plateau. In addition, we tested the relationship between plant diversity and spatial stability of grassland productivity. Results showed that an increase in plant diversity significantly enhanced community productivity and its standard deviation, while reducing the coefficient of variation in productivity. The influence of plant diversity on productivity and the reciprocal of productivity variability coefficient was not affected by vegetation types. The absolute values of the regression slopes between climate factors and productivity in alpine meadow communities with higher plant diversity were smaller than those in alpine meadow communities with lower plant diversity. In other words, alpine meadow communities with higher plant diversity exhibited a weaker response to climatic factors in terms of productivity, whereas those with lower plant diversity showed a stronger response. Our results indicate that high plant diversity buffers the impact of ambient pressure (e.g., precipitation, temperature) on alpine meadow productivity, and significantly enhanced the spatial stability of grassland productivity. This finding provides a theoretical basis for maintaining the stability of grassland ecosystems and scientifically managing alpine grasslands under the continuous climate change.

Machine Learning for Experimental Reactivity of a Set of Metal Clusters toward C-H Activation.

Understanding the mechanisms of C-H activation of alkanes is a very important research topic. The reactions of metal clusters with alkanes have been extensively studied to reveal the electronic features governing C-H activation, while the experimental cluster reactivity was qualitatively interpreted case by case in the literature. Herein, we prepared and mass-selected over 100 rhodium-based clusters (RhVO and RhCoO) to react with light alkanes, enabling the determination of reaction rate constants spanning six orders of magnitude. A satisfactory model being able to quantitatively describe the rate data in terms of multiple cluster electronic features (average electron occupancy of valence s orbitals, the minimum natural charge on the metal atom, cluster polarizability, and energy gap involved in the agostic interaction) has been constructed through a machine learning approach. This study demonstrates that the general mechanisms governing the very important process of C-H activation by diverse metal centers can be discovered by interpreting experimental data with artificial intelligence.
