Optimizing low-protein diets with edible dock powder: Integrated effects on growth performance, slaughter quality, Organ weights, Muscle quality, and Cecal microbiota in growing Sanhua geese.
This study evaluated the effects of supplementing low-protein diets with Edible Dock Powder (EDP) on the growth performance, slaughter traits, serum biochemical parameters, muscle quality, and cecal microbiota of Sanhua geese. A total of 288 healthy, five-week-old Sanhua geese were randomly assigned to six dietary treatments in a 3 × 2 factorial design, with three crude protein levels (16.00 %, 14.50 %, and 13.00 %) and two levels of EDP supplementation (0 % and 2.50 %). Two-way ANOVA and Duncan's multiple range test were used for statistical analysis. EDP supplementation significantly increased average daily gain (ADG) and improved feed-to-gain ratio (F/G) during both growth phases (P<0.01). Lower protein levels significantly reduced average daily feed intake (ADFI) and increased the apparent digestibility of gross energy (ADGE) (P<0.01). EDP significantly improved slaughter rate and eviscerated yield (P<0.05), while reducing liver weight and webbed feet yield (P<0.01). Reduced protein levels decreased serum globulin (GLB) and increased blood urea nitrogen (BUN) levels (P<0.05), with significant interactions between protein levels and EDP supplementation (P<0.05). EDP also significantly altered the cecal microbiota composition, reducing the relative abundance of Actinobacteria, Megamonas, and Collinsella (P<0.05), and affecting KEGG pathways related to protein modification and secondary metabolite degradation (P<0.05). Identification of Plant Peroxidases Catalyzing the Degradation of Fluorinated Aromatics Using a Peroxidase Library Approach.
In this work, the degradation of mono- and polyfluorinated phenolic compounds was demonstrated by a series of crude plant peroxidases, including horseradish root (HRP) and six members of the genus. Highly active samples were identified using a library screening approach in which more than 50 crude plant samples were initially evaluated for defluorination activity toward 4-fluorophenol. The highest concentrations were observed in the HRP, pumpkin skin (PKS), and butternut squash skin (BNS), which consistently gave the highest intrinsic rates of decomposition for all the substrates tested. Although HRP exhibited a significant decrease in activity with increased fluorination of the phenolic substrate, PKS showed only minor reductions. Furthermore, in silico studies indicated that the active site of HRP poorly accommodates the steric bulk of additional fluorines, causing the substrate to dock farther from the catalytic heme and thus slowing the catalysis rate. We propose that the PKS active site might be larger, allowing closer access to the perfluorinated substrate, and therefore maintaining higher activity compared to the HRP enzyme. However, detailed kinetic characterization studies of the peroxidases are recommended. Conclusively, the high catalytic activity of PKS and its high yield per gram of tissue make it an excellent candidate for developing environmentally friendly biocatalytic methods for degrading fluorinated aromatics. AI-Enabled Ultra-large Virtual Screening Identifies Potential Inhibitors of Choline Acetyltransferase for Theranostic Purposes.
Alzheimer's disease (AD) and related dementias are among the primary neurological disorders and call for the urgent need for early-stage diagnosis to gain an upper edge in therapeutic intervention and increase the overall success rate. Choline acetyltransferase (ChAT) is the key acetylcholine (ACh) biosynthesizing enzyme and a legitimate target for the development of biomarkers for early-stage diagnosis and monitoring of therapeutic responses. It is also a theranostic target for tackling colon and lung cancers, where overexpression of non-neuronal ChAT leads to the production of acetylcholine, which acts as an autocrine growth factor for cancer cells. Theranostics is a hybrid of diagnostics and therapeutics that can be used to locate cancer cells using radiotracers and kill them without affecting other healthy tissues. Traditional virtual screening protocols have a lot of limitations; given the current rate of chemical database expansion exceeding billions, much faster screening protocols are required. Deep docking (DD) is one such platform that leverages the power of deep neural network (DNN)-based virtual screening, empowering researchers to dock billions of molecules in a speedy, yet explicit manner. Here, we have screened 1.3 billion compounds library from the ZINC20 database, identifying the best-performing hits. With each iteration run where the first iteration gave ∼116 million hits, the second iteration gave ∼3.7 million hits, and the final third iteration gave 168,447 hits from which further refinement gave us the top 5 compounds as potential ChAT inhibitors. Using artificial intelligence (AI) to assess the prevalence of false or misleading health-related claims.
Complementary healthcare in the United Kingdom is subject to voluntary, publicly funded regulation. Many such practices include pseudoscience. There are concerns that regulated practitioners make misleading health claims. This study used an artificial intelligence (AI) tool to measure the prevalence of such claims. Websites operated by practitioners of pseudoscientific complementary and alternative medicine, registered with the Complementary and Natural Healthcare Council, were downloaded and assessed by the AI, which determined whether a website was relevant to the investigation and, if so, identified health-related claims that it judged as false or misleading, supplying a rationale. Of 6096 registrants, 1326 met the selection criteria, of which 872 clinics had 725 relevant and operational websites. The AI assessed text from 11 771 web pages, identifying false or misleading claims in 704 (97%) of the websites. The AI's performance was quality-assured by four human assessors, who manually reviewed 23 relevant web pages. Humans identified on average 39.5 claims likely to be judged false or misleading by advertising regulators, the AI identified 36. Humans misidentified an average of 4.8 claims, AI misidentified two. Most practitioners of pseudoscientific therapies registered with the Complementary and Natural Healthcare Council make misleading health claims online. Assessing small molecule conformational sampling methods in molecular docking.
Small molecule conformational sampling plays a pivotal role in molecular docking. Recent advancements have led to the emergence of various conformational sampling methods, each employing distinct algorithms. This study investigates the impact of different small molecule conformational sampling methods in molecular docking using UCSF DOCK 3.7. Specifically, six traditional sampling methods (Omega, BCL::Conf, CCDC Conformer Generator, ConfGenX, Conformator, RDKit ETKDGv3) and a deep learning-based model (Torsional Diffusion) for generating conformational ensembles are evaluated. These ensembles are subsequently docked against the Platinum Diverse Dataset, the PoseBusters dataset and the DUDE-Z dataset to assess binding pose reproducibility and screening power. Notably, different sampling methods exhibit varying performance due to their unique preferences, such as dihedral angle sampling ranges on rotatable bonds. Spatial and seasonal variations in trace metals in marine sediments from the Dubai coastal environment.
This study was conducted to assess sediment trace metals (Cd, Cr, Cu, Pb, Ni and Zn) contamination using a systematic approach by collecting sediment samples from 8 transects along the Dubai coastline, each 10 km long, and each transect included its nearshore sediment sampling station. Additionally, 10 sediment samples were collected from the Dubai creek and other potential sources of metal pollution. The sediment samples were collected in December and again in August. However, no significant difference in sediment metal concentration was found between the two sampling campaigns. The sediment trace metal concentrations (0.92-1.31 mg Cd/kg, 2.82-176.6 mg Cr/kg, 2.27-621.67 mg Cu/kg, 0.88-23.6 mg Pb/kg, 1.92-192.2 mg Ni/kg and 9.1-391.05 mg Zn/kg) showed considerable variability, except for Cd (1.08 ± 0.06 mg/kg, 5.55% variability). Despite this, no significant differences in sediment metal concentrations were found between the sampled transects. However, significant variations in Cr, Cu, Pb and Zn were evident between distances from the shoreline to offshore stations along the Dubai coast, and the nearshore locations presented clear evidence of elevated/maximum sediment metal concentrations. Most of the sediment trace metal concentrations, however, were found well within the sediment quality guidelines (SQGs) for nearshore sediments. Trace metal contamination hotspots, nonetheless, were identified at some nearshore stations as determined by metal level exceedance over the SQGs, background levels and the pollution load index, with limited potential ecological risk. Overall, the findings suggest that sediments in the Dubai coastal environment are mainly influenced by anthropogenic activities in stations located in the proximity of ship maintenance, ports, and industrial areas such as Dry Dock, Jaddaf, Jebel Ali Port, Wharfage, Hamriya and DUBAL.Hamdan Al Shaer, Peter S Hooda, Shahid Mustafa, Laila A Mohamed
