Novel class of peptides disintegrating biological membranes to aid in the characterization of membrane proteins.
Styrene-maleic acid (SMA) and similar amphiphilic copolymers are known to cut biological membranes into lipid nanoparticles/nanodiscs containing membrane proteins apparently in their relatively native membrane lipid environment. Our previous work demonstrated that membrane raft microdomains resist such disintegration by SMA. The use of SMA in studying membrane proteins is limited by its heterogeneity and the inability to prepare defined derivatives. In the present paper, we demonstrate that some amphiphilic peptides structurally mimicking SMA also similarly disintegrate cell membranes. In contrast to the previously used copolymers, the simple peptides are structurally homogeneous. We found that their membrane disintegrating activity increases with their length (reaching optimum at 24 amino acids) and requires a basic primary structure, i.e. (XXD)n, where X represents a hydrophobic amino acid (optimally phenylalanine), D aspartic acid and n is the number of repeats of these triplets. These peptides may provide opportunities for various well-defined potentially useful modifications in the study of membrane protein biochemistry. A Pilot Study In Fatigue Risk Management amongst Obstetrics and Gynecology Residents at the University of Alberta.
La lutte contre la fatigue est un défi constant dans les programmes de résidence en chirurgie, car la fatigue pèse autant sur la qualité des soins que sur le bien-être des résidents. Les nuances de la fatigue constante sont mal comprises, surtout lorsqu'elle se rapporte aux périodes de garde. Cette étude pilote vise à explorer l'ampleur du problème chez les résidents en obstétrique et gynécologie et à cerner les obstacles à l'obtention d'un repos adéquat. MéTHODE: Un sondage a été envoyé à tous les résidents en gynécologie et obstétrique de l'Université de l'Alberta afin de recueillir des données démographiques et des informations de référence sur le sommeil, en plus d'évaluer les obstacles au sommeil perçus. Les résidents volontaires ont ensuite participé à la deuxième partie de l'étude, qui consistait à consigner tous les appels reçus lors d'une garde de 12 heures et à leur attribuer une cote d'acuité. Des méthodes mixtes ont été utilisées, notamment une analyse thématique pour le sondage sur l'acuité et des statistiques descriptives pour le sondage initial. RéSULTATS: Au total, 21 résidents ont répondu au sondage initial (67,7 %) et 17 gardes de 12 heures ont été consignées. Les résidents juniors (R1-R2) et les porteurs d'un téléavertisseur pour les cas à faible risque dormaient moins lorsqu'ils étaient de garde, et ce, même s'ils ne recevaient aucun appel. Toutefois, les obstacles au sommeil n'étaient pas différents chez les résidents en fin de formation et les porteurs d'un téléavertisseur pour les cas à risque élevé. Bien que les facteurs de contribution à la fatigue étaient différents entre les gardes à faible risque et à risque élevé (volume et acuité, respectivement), les deux groupes attribuaient la fatigue aux appels non urgents. À l'examen des appels, 49,4 % ont été considérés comme non urgents. Liquid-Phase Cyclic Chemiluminescence for the Identification of Cobalt Speciation.
Accurate discrimination of metal species is a significant analytical challenge. Herein, we propose a novel methodology based on liquid-phase cyclic chemiluminescence (CCL) for the identification of cobalt speciation. The CCL multistage signals () of the luminol-HO reaction catalyzed by different cobalt species have different decay coefficients . Thereby, we can facilely identify various cobalt species according to the distinguishable values, including the complicated and structurally similar cobalt complexes, such as analogues of [Co(NH)X] (X = Cl, HO, and NH), Co(II) porphyrins, and bis(2,4-pentanedione) cobalt(II) derivatives. Especially, the number of substituent atoms also influences the value greatly, which allows excellent discrimination between complexes that only have a subtle difference in the substituent group. In addition, linear discriminant analysis based on provides a complementary solution to improve the differentiating ability. We performed density functional theory calculations to investigate the interaction mode of HO over cobalt species. A close negative correlation between the adsorption energy and the value is observed. Moreover, the calculation of energy evolutions of HO decomposition into a double hydroxide radical shows that a high level of consistency exists between the activation energy barrier and the value. The results further demonstrate that the decay coefficient of the CCL multistage signal is associated with the catalytic reactivity of the cobalt species. Single nucleotide polymorphisms (SNPs) in the open reading frame (ORF) of prion protein gene (PRNP) in Nigerian livestock species.
Prion diseases, also known as transmissible spongiform encephalopathies (TSEs) remain one of the deleterious disorders, which have affected several animal species. Polymorphism of the prion protein (PRNP) gene majorly determines the susceptibility of animals to TSEs. However, only limited studies have examined the variation in PRNP gene in different Nigerian livestock species. Thus, this study aimed to identify the polymorphism of PRNP gene in Nigerian livestock species (including camel, dog, horse, goat, and sheep). Monitoring Off-Shore Fishing in the Northern Indian Ocean Based on Satellite Automatic Identification System and Remote Sensing Data.
Satellite-derived Sea Surface Temperature (SST) and sea-surface Chlorophyll a concentration (Chl-a), along with Automatic Identification System (AIS) data of fishing vessels, were used in the examination of the correlation between fishing operations and oceanographic factors within the northern Indian Ocean from March 2020 to February 2023. Frequency analysis and the empirical cumulative distribution function (ECDF) were used to calculate the optimum ranges of two oceanographic factors for fishing operations. The results revealed a substantial influence of the northeast and southwest monsoons significantly impacting fishing operations in the northern Indian Ocean, with extensive and active operations during the period from October to March and a notable reduction from April to September. Spatially, fishing vessels were mainly concentrated between 20° N and 6° S, extending from west of 90° E to the eastern coast of Africa. Observable seasonal variations in the distribution of fishing vessels were observed in the central and southeastern Arabian Sea, along with its adjacent high sea of the Indian Ocean. Concerning the marine environment, it was observed that during the northeast monsoon, the suitable SST contributed to high CPUEs in fishing operation areas. Fishing vessels were widely distributed in the areas with both mid-range and low-range Chl-a concentrations, with a small part distributed in high-concentration areas. Moreover, the monthly numbers of fishing vessels showed seasonal fluctuations between March 2020 and February 2023, displaying a periodic pattern with an overall increasing trend. The total number of fishing vessels decreased due to the impact of the COVID-19 pandemic in 2020, but this was followed by a gradual recovery in the subsequent two years. For fishing operations in the northern Indian Ocean, the optimum ranges for SST and Chl-a concentration were 27.96 to 29.47 °C and 0.03 to 1.81 mg/m, respectively. The preliminary findings of this study revealed the spatial-temporal distribution characteristics of fishing vessels in the northern Indian Ocean and the suitable ranges of SST and Chl-a concentration for fishing operations. Metabolic Changes in Serum and Milk of Holstein Cows in Their First to Fourth Parity Revealed by Biochemical Analysis and Untargeted Metabolomics.
The performance of dairy cows is closely tied to the metabolic state, and this performance varies depending on the number of times the cows have given birth. However, there is still a lack of research on the relationship between the metabolic state of Holstein cows and the performance of lactation across multiple parities. In this study, biochemical analyses and metabolomics studies were performed on the serum and milk from Holstein cows of parities 1-4 (H1, N = 10; H2, N = 7; H3, N = 9; H4, N = 9) in mid-lactation (DIM of 141 ± 4 days) to investigate the link between performance and metabolic changes. The results of the milk quality analysis showed that the lactose levels were highest in H1 ( = 0.036). The total protein content in the serum increased with increasing parity ( = 0.013). Additionally, the lipase activity was found to be lowest in H1 ( = 0.022). There was no difference in the composition of the hydrolyzed amino acids in the milk among H1 to H4. However, the free amino acids histidine and glutamate in the serum were lowest in H1 and highest in H3 ( < 0.001), while glycine was higher in H4 ( = 0.031). The metabolomics analysis revealed that 53 and 118 differential metabolites were identified in the milk and serum, respectively. The differential metabolites in the cows' milk were classified into seven categories based on KEGG. Most of the differential metabolites in the cows' milk were found to be more abundant in H1, and these metabolites were enriched in two impact pathways. The differential metabolites in the serum could be classified into nine categories and enriched in six metabolic pathways. A total of six shared metabolites were identified in the serum and milk, among which cholesterol and citric acid were closely related to amino acid metabolism in the serum. These findings indicate a significant influence of blood metabolites on the energy and amino acid metabolism during the milk production process in the Holstein cows across 1-4 lactations, and that an in-depth understanding of the metabolic changes that occur in Holstein cows during different lactations is essential for precision farming, and that it is worthwhile to further investigate these key metabolites that have an impact through controlled experiments.Zixin Liu, Aoyu Jiang, Xiaokang Lv, Chuanshe Zhou, Zhiliang Tan

The application of high-throughput sequencing (HTS) technologies has revolutionized research on phytoplankton biodiversity by generating an unprecedented amount of molecular data in marine ecosystem surveys. However, high-level of molecular diversity uncovered in HTS-based metabarcoding analyses may lead to overinterpretation of phytoplankton diversity due to excessive intra-genomic variations (IGVs). The aims in this study are to explore the nature of phytoplankton molecular diversity and to test the hypothesis. We carried out single-cell metabarcoding analysis of 18S rDNA V4 sequences obtained in single Noctiluca scintillans cells isolated from various sites in coastal waters of China. Results showed that each single N. scintillans cell harbored a high level of IGVs with about 100 amplicon sequence variants (ASVs). The large numbers of non-dominant ASVs identified in N. scintillans cells, which might correspond to the larger numbers of ASVs annotated as N. scintillans and showed similar temporal dynamics in metabarcoding analyses, could inflate the inter-species diversity or intra-species genetic diversity. In addition, there were large numbers of additional ASVs that were not annotated as N. scintillans. These non-N. scintillans ASVs might represent diverse preys for N. scintillans, consistent with previous reports that N. scintillans may act as chance predator of a broad-spectrum preys. This single-cell study has unambiguously demonstrated that the existence of high levels of IGVs in N. scintillans and most likely many other phytoplankton species, demonstrating that the majority of the molecular diversity revealed in metabarcoding analysis, which were generally interpreted as the sum of inter-species diversity and intra-species diversity, actually included high levels of IGVs and should be interpreted with caution.Kuiyan Liu, Xianliang Huang, Xiangxiang Ding, Nansheng Chen
