Minimal Clinically Important Differences in Functional Motor Scores in Adult Patients with Spinal Muscular Atrophy.

In patients with spinal muscular atrophy (SMA), functional disease scores are frequently used to evaluate the course of the disease and the efficacy of treatment. The aim of our study was to propose minimal clinically important difference (MCID) values for motor scores in order to estimate the degree of change within a functional score that can be considered clinically meaningful.

Role of human papillomavirus status after conization for high-grade cervical intraepithelial neoplasia.

Human papillomavirus (HPV) is the well-established etiologic factor for cervical neoplasia. Cervical conization constitutes an effective treatment for high-grade cervical intraepithelial neoplasia (HG-CIN). We conducted an observational study for long-term outcomes and HPV genotype changes after conization for HG-CIN. Between 2008 and 2014, patients with newly diagnosed HG-CIN before conization (Surveillance New [SN] group) and those who had undergone conization without hysterectomy (Surveillance Previous [SP] group) were enrolled. HPV testing and Pap smear were performed periodically for the SN and SP (collectively S) groups. All other patients receiving conization for HG-CIN during the study period were identified from our hospital database. Those eligible but not enrolled into this study were assigned to the non-Surveillance (non-S) group. For the S group (n = 493), the median follow-up period was 74.3 months. Eighty-four cases had recurrent CIN grade 2 or worse (CIN2+) (5-year cumulative rate: 14.8%), of which 6 had invasive cancer. Among the 84 patients, 65 (77.4%) exhibited type-specific persistence in the paired HPV results, whereas only 7 (8.3%) harbored new HPV types that belonged to the 9-valent vaccine types. Among the 7397 non-S patients, 789 demonstrated recurrent CIN2+, of which 57 had invasive cancer. The stages distribution of those progressed to invasive cancer in the non-S group were more advanced than the S group (P = 0.033). Active surveillance might reduce the severity of those progressed to cancer. Because a majority of the patients with recurrent CIN2+ had persistent type-specific HPV infections, effective therapeutic vaccines are an unmet medical need.

An unbiased molecular approach using 3'UTRs resolves the avian family-level tree of life.

Presumably, due to a rapid early diversification, major parts of the higher-level phylogeny of birds are still resolved controversially in different analyses or are considered unresolvable. To address this problem, we produced an avian tree of life, which includes molecular sequences of one or several species of ∼ 90% of the currently recognized family-level taxa (429 species, 379 genera) including all 106 for the non-passerines and 115 for the passerines (Passeriformes). The unconstrained analyses of noncoding 3-prime untranslated region (3'UTR) sequences and those of coding sequences yielded different trees. In contrast to the coding sequences, the 3'UTR sequences resulted in a well-resolved and stable tree topology. The 3'UTR contained, unexpectedly, transcription factor binding motifs that were specific for different higher-level taxa. In this tree, grebes and flamingos are the sister clade of all other Neoaves, which are subdivided into five major clades. All non-passerine taxa were placed with robust statistical support including the long-time enigmatic hoatzin (Opisthocomiformes), which was found being the sister taxon of the Caprimulgiformes. The comparatively late radiation of family-level clades of the songbirds (oscine Passeriformes) contrasts with the attenuated diversification of non-passeriform taxa since the early Miocene. This correlates with the evolution of vocal production learning, an important speciation factor, which is ancestral for songbirds and evolved convergent only in hummingbirds and parrots. Since 3'UTR-based phylotranscriptomics resolved the avian family-level tree of life, we suggest that this procedure will also resolve the all-species avian tree of life.

Giant magnetoresistance and dual spin filtering effect in ferromagnetic 6,6,12/γ-graphyne zigzag nanoribbon lateral heterojunction.

Based on non-equilibrium Green's function combined with density functional theory (NEGF-DFT), we investigate the spin dependent transport in the ferromagnetic 6,6,12/γ-graphyne zigzag nanoribbon (GYZNR) heterojunction under different magnetic configurations. It is found that, at low bias ([-0.05, 0.1] V), the junction presents metallic transport with negligible spin polarization in parallel configuration (PC) while it behaves as an insulator in anti-parallel configuration (APC), which results in giant magnetoresistance. Interestingly, when we increase the bias voltage beyond [-0.05, 0.1] V, dual spin filtering characterized by electron transport of different spin channels under different polarity of bias is observed in APC but not in PC. All these findings are understood from the symmetry matching of wave functions in two nanoribbons at equilibrium or finite bias. Furthermore, dual spin filtering can also be achieved in PC by applying a gate voltage on the central interface region, which arises from the shift of different single spin channel of the central gate region into the bias window at a different polarity of the gate voltage. Thus, our work demonstrates the great potential of the 6,6,12/γ-GYZNR heterojunction as a multi-functional device and its great perspectives in carbon-based nanoelectronics and spintronics.

Transient coarsening and the motility of optically heated Janus colloids in a binary liquid mixture.

A gold-capped Janus particle suspended in a near-critical binary liquid mixture can self-propel under illumination. We have immobilized such a particle in a narrow channel and carried out a combined experimental and theoretical study of the non-equilibrium dynamics of a binary solvent around it - lasting from the very moment of switching illumination on until the steady state is reached. In the theoretical study we use both a purely diffusive and a hydrodynamic model, which we solve numerically. Our results demonstrate a remarkable complexity of the time evolution of the concentration field around the colloid. This evolution is governed by the combined effects of the temperature gradient and the wettability, and crucially depends on whether the colloid is free to move or is trapped. For the trapped colloid, all approaches indicate that the early time dynamics is purely diffusive and characterized by composition layers travelling with constant speed from the surface of the colloid into the bulk of the solvent. Subsequently, hydrodynamic effects set in. Anomalously large nonequilibrium fluctuations, which result from the temperature gradient and the vicinity of the critical point of the binary liquid mixture, give rise to strong concentration fluctuations in the solvent and to permanently changing coarsening patterns not observed for a mobile particle. The early time dynamics around initially still Janus colloids produces a force which is able to set the Janus colloid into motion. The propulsion due to this transient dynamics is in the direction opposite to that observed after the steady state is attained.

Expanding the applicability of cytochrome P450s and other haemoproteins.

Cytochrome P450BM3 has long been regarded as a promising candidate for use as a biocatalyst, owing to its excellent efficiency for the hydroxylation of unactivated C-H bonds. However, owing because of its high substrate specificity, its possible applications have been severely limited. Consequently, various approaches have been proposed to overcome the enzyme's natural limitations, thereby expanding its substrate scope to encompass non-native substrates, evoking chemoselectivity, regioselectivity and stereoselectivity and enabling previously inaccessible chemical conversions. Herein, these approaches will be classified into three categories: (1) mutagenesis including directed evolution, (2) haem substitution with artificial cofactors and (3) use of substrate mimics, 'decoy molecules'. Herein, we highlight the representative work that has been conducted in above three categories for discussion of the future outlook of P450BM3 in green chemistry.

Sonochemical synthesis of aluminium and aluminium hybrids for remediation of toxic metals.

Spherical shaped nano-size aluminium oxide and its hybrids with indole and indole derivatives have been synthesized using sol-gel and post grafting methods coupled with sonication (Branson Digital SonifierS-250D; 20 kHz; 40%) for the remediation of toxic metals (lead and mercury). Different spectroscopic techniques (FTIR, SEM, BET, XRD, and XPS) have been applied to assess the properties of synthesized aluminium oxide and its hybrids. FTIR spectra showed the absorption bands of aluminium oxide (Al-O-Al) and aluminium hybrids (Al-O-C) at 800-400 cm and 1650-1100 cm region, respectively. SEM showed spherical shaped clusters of aluminium oxide which changed into the net-shape structure after the hybrid synthesis. It is worth noting that sonication energy increases the total surface area of aluminium oxide when it gets hybridized with indole and its derivatives from 82 m/g to 167 m/g; it also improved the product yield from 68% to 78%. Simultaneously, FTIR, SEM and BET analysis of non-sonicated aluminium oxide and its hybrids were also recorded for comparison. While XRD and XPS analysis were only conducted for sonicated aluminium oxide and its hybrids to manifest the structural and compositional properties. XRD patterns indexed as the cubic crystal system with an average 41 nm crystallite size of sonicated aluminium oxide which remains unaffected after hybrid synthesis. A survey scan under XPS confirmed the presence of all expected elements (aluminium, oxygen, carbon, nitrogen) and deconvolution of each recorded peak showed binding of element with its neighboring elements. The performance of aluminium oxide and its hybrids synthesize with and without sonication are also evaluated using a time-dependent batch adsorption protocol optimize for one hour. The maximum adsorption of lead (37%) and mercury (40%) are found onto sonicated aluminium oxide. The sonicated aluminium hybrids showed 43-63% of lead and 55-67% of mercury at pH 7. The fitness of experimental data using adsorption kinetics and isotherms revealed that adsorption follows Pseudo-second-order kinetic, Langmuir, and Freundlich isotherms.

Influence of surface modification of Ti6Al7Nb alloy on adhesion of poly (lactide-co-glycolide) coating (PLGA).

The paper presents the results of studies on the influence of surface topography and wettability of a Ti6Al7Nb alloy substrate on the adhesion of a PLGA coating. The alloy surface was modified using a mechanical pre-treatment including: grinding, vibration treatment, mechanical polishing, sandblasting and anodic oxidation. The polymer coating was applied to the metal substrate by a dip-coating method. The scope of the research included microscopic observations of the substrate and polymer coating using SEM and acoustic microscopy. In addition, studies on the wettability and topography of the polymer coating and the metal substrate, the thickness of the polymer coating as well as qualitative and quantitative testing of the adhesion of the polymer coating to the substrate were carried out. Coating adhesion tests were conducted for samples in the initial state and after 6 weeks exposure to Ringer's solution. Analysis of the results indicates the influence of the method used to modify the metal substrate on its topography and wettability. These parameters affect the thickness of the obtained polymer coating. Regardless the parameters of the metal substrate, a qualitative analysis of the adhesion of the coating applied to the substrate of the Ti6Al7Nb alloy did not show any delamination for both samples exposed and non-exposed to Ringer's solution. On the other hand, quantitative scratch-test studies showed different adhesion of the polymer coating to the substrate depending on the surface topography obtained by various modification methods. The cytotoxicity test conducted by the indirect method using extracts confirmed that the surface modification does not affect cell growth. The complex methods of surface pre-treatment of the alloy together with the kind of polymer selected for the study allowed to develop well adhered PLGA layers on Ti6Al7Nb intended for short term implants. The lack of delamination of the layer during 6 weeks was proved, what allows for maintaining the protection function of the layer during this period and contribute to improving biocompatibility.
