Complex cases of multiple allergies can be particularly difficult to diagnose using standard methods such as skin prick tests and assessment of a patient's allergic history. Multiplex allergy testing may improve outcomes for allergy patients by avoiding misdiagnosis and providing reassurance. The ImmunoCAP Immuno Solid-Phase Allergen Chip (ISAC) 112 is a CE-marked, molecular, multiplex, allergy test that can test for IgE antibodies to 112 components from 51 allergen sources. However, its clinical utility is unknown and is difficult to estimate due to the complexity of the diagnostic pathway in which it is used.

Ultrastructure of the intestinal system in unfed larvae of Limnesia maculata (O.F. Müller, 1776) (Acariformes, Limnesiidae).

The intestinal system of unfed fresh-water mite larvae Limnesia maculata (O.F. Müller, 1776) (Acariformes, Limnesiidae) has been studied with transmission electron microscopy. The intestinal system is composed of the foregut, including the pharynx and the esophagus, the sac-like blind midgut and the excretory organ. The pharynx begins with the mouth covered by the labrum. The pharynx runs along the bottom of the gnathosoma and shows ventral dilators. No valves are expressed between the pharynx and the esophagus. The esophagus possesses strongly plicate walls and, before entering the midgut, passes through the brain. The sac-like midgut does not reveal a well-pronounced lumen, developed epithelium and separate lobes. It consists of the two cell types of endoderm origin mixed in the midgut volume. The first type - the vacuolated cells - does not possess Golgi bodies and lysosomal apparatus and shows electron-lucent vacuoles with a granular inclusion inside. These cells apparently do not take part in digestion of the embryonic yolk. The second type - the non-vacuolated cells - shows both, a well developed Golgi complex and large heterolysosomes, and obviously digests the embryonic yolk. Consequently, they may be attributed as specialized vitellophages. Nevertheless, both cell types may take part in formation of the definitive midgut epithelium. The sac-like thin-walled excretory organ is strongly dilated and contains the embryonic wastes in the form of electron-dense globules and birefringent particles. No muscle envelope surrounds the excretory organ. The embryonic wastes together with wastes accumulated during feeding may be evacuated from the organ only after completion of feeding. The excretory canal on this developmental stage is not connected with the excretory organ. It opens to the outside with a simple slit-like excretory pore. Before feeding, larvae have to pass the process of the post-molt development before their midgut would be ready to receive nutrients.

Glucose 6-phosphate dehydrogenase inhibition sensitizes melanoma cells to metformin treatment.

Most cancer cells exacerbate the pentose phosphate pathway (PPP) to enhance biosynthetic precursors and antioxidant defenses. Metformin, which is used as a first-line oral drug for the treatment of type 2 diabetes, has been proposed to inhibit the malignant progression of different types of cancers. However, metformin has shown poor efficacy as single agent in several clinical trials. Thus, the aim of the present work was to investigate whether the pharmacological inhibition of G6PDH, the first and rate-limiting enzyme of the PPP, by 6-amino nicotinamide (6-AN) potentiates the antitumoral activity of metformin on different human melanoma cell lines. Our results showed that 6-AN has sensitizing properties to metformin cytotoxicity. The combination of metformin and 6-AN decreased glucose consumption and lactate production, altered the mitochondrial potential and redox balance, and thereby blocked melanoma cell progression, directing cells to apoptosis and necrosis. To our knowledge, this is the first study describing the effect of this combination. Future preclinical studies should be performed to reveal the biological relevance of this finding.

An integrated approach to uncover quality markers of Traditional Chinese medicine underlying chemical profiling, network target selection and metabolomics approach: Guan-Xin-Jing capsule as a model.

The "multi-component and multi-target" property of traditional Chinese medicine (TCM) makes the quality control more difficult, which remains a key concern hindering the space of progress in its modernization and globalization. This study aims to provide a new strategy to discover quality markers related to the efficacy and stability of TCM based on the bioactive markers finding and differential markers finding, Guan-Xin-Jing capsule (GXJC) was used as a model. Firstly, the UHPLC-QTOF-MS/MS was performed to identify the chemical profiling of GXJC and 148 compounds were unequivocally or tentatively identified. Subsequently, the compounds with relatively low intensity, low oral bioavailability and unsuited drug likeness were removed. In addition, a network based approach by integrating candidate compound genes and disease genes was applied to further filter out the potential active compounds against coronary heart disease (CHD) and finally 7 compounds were identified as the potential bioactive markers. Moreover, to ensure the batch-to-batch consistency of GXJC, an untargeted UHPLC-QTOF-MS/MS based metabolomics approach and multivariate statistical analysis was performed and 5 compounds were identified as the potential differential markers to illustrate the discrepancy among 27 batches of GXJC. Using these strategies above, a total of 11 chemical compounds in GXJC were screened out as quality markers(Q-markers). The information generated with our strategy should be more helpful to guide the selection of the Q-markers and lay the foundation for quality control of TCM.

A comprehensive investigation of industrial plastic pellets on beaches across the Laurentian Great Lakes and the factors governing their distribution.

Industrial, pre-consumer pellets are a major type of plastics pollution found on shorelines worldwide. This study investigates the distribution and characteristics of plastic pellets accumulated on beaches of the Laurentian Great Lakes of North America and provides a "snapshot" of pellet distribution in a lake system that accounts for 21% of the world's freshwater reserves. We sampled pellets simultaneously from 10m quadrats on 66 beaches and characterized the 12,595 pellets collected (average of 19.1 pellets/m). Forty-two beaches contained pellets and 86% of the pellets were found on three beaches: Rossport (Lake Superior), Baxter (Lake Huron), and Bronte (Lake Ontario). The number of pellets on each beach was compared with factors hypothesized to control their accumulation. In general, positive correlations were found between pellet abundance and watershed population, number of plastic-related industries, and proximity to a river mouth, although for Lake Superior, abundance was related to a train spill that took place over 10 years ago. Beach grain size appears to be related to pellet abundance, with very fine sand, fine sand and medium sand containing the greatest number of pellets. All pellets were visually characterized based on size, color, shape, weathering, and distinguishing traits. The predominant color was white, oblate shapes were most common, and the main distinguishing trait was a dimple. Most pellets showed little evidence of weathering, with the weathered samples mainly from Lakes Erie and Ontario. Lake Ontario pellets were the most varied, with 6/7 shapes, 35/40 colors, and 21/25 distinguishing traits, indicating a wider range of pellet sources compared to the other lakes. Polymer compositions were mainly polyethylene (PE) and polypropylene (PP). Our results will lead to increased recognition of regional pellet pollution in the Great Lakes watershed, thereby motivating change during their production, transport and use.
