Temporal expectation driven by rhythmic cues compared to that driven by symbolic cues provides a more precise attentional focus in time.

Temporal expectation is the ability to select the precise point in time for doing something to produce an optimal effect. Two sources of information that humans use to generate temporal expectations are rhythmic and symbolic cues. Both types of cues have been proven effective in directing attention to a future point in time resulting in improved performance. However, the temporal precision of the two forms of temporal expectation have rarely been compared. In the current study, 17 participants performed two temporal expectation tasks in which either a rhythmic cue or a symbolic cue indicated that a future target would appear after a 500-ms (short) or 1,500-ms (long) interval; the target appeared at the expected time in 54% of trials and at an unexpected earlier or later interval in 36% of trials. In both tasks, we observed that the reaction time (RT) curves were U-shaped, with a slower RT for the earlier and later unexpected intervals and a faster RT for intervals approaching the expected point in time. Furthermore, we found a significant interaction between task and the quadratic term of temporal expectation, which indicates that the U-shaped RT curves for the rhythmic cue task are steeper than those for the symbolic cue task. Thus, the current results revealed that compared with symbolic cues, temporal expectation driven by rhythmic cues provides a more precise attentional focus in time.

Tutorial: design and fabrication of nanoparticle-based lateral-flow immunoassays.

Lateral-flow assays (LFAs) are quick, simple and cheap assays to analyze various samples at the point of care or in the field, making them one of the most widespread biosensors currently available. They have been successfully employed for the detection of a myriad of different targets (ranging from atoms up to whole cells) in all type of samples (including water, blood, foodstuff and environmental samples). Their operation relies on the capillary flow of the sample throughout a series of sequential pads, each with different functionalities aiming to generate a signal to indicate the absence/presence (and, in some cases, the concentration) of the analyte of interest. To have a user-friendly operation, their development requires the optimization of multiple, interconnected parameters that may overwhelm new developers. In this tutorial, we provide the readers with: (i) the basic knowledge to understand the principles governing an LFA and to take informed decisions during lateral flow strip design and fabrication, (ii) a roadmap for optimal LFA development independent of the specific application, (iii) a step-by-step example procedure for the assembly and operation of an LF strip for the detection of human IgG and (iv) an extensive troubleshooting section addressing the most frequent issues in designing, assembling and using LFAs. By changing only the receptors, the provided example procedure can easily be adapted for cost-efficient detection of a broad variety of targets.

A causal role for the cerebellum in semantic integration: a transcranial magnetic stimulation study.

Mounting evidence suggests that the cerebellum, a structure previously linked to motor function, is also involved in a wide range of non-motor processes. It has been proposed that the cerebellum performs the same computational processes in both motor and non-motor domains. Within motor functions, the cerebellum is involved in the integration of signals from multiple systems. Here we hypothesized that cerebellum may be involved in integration within semantic memory as well. Specifically, understanding a noun-adjective combination (e.g. red apple) requires combining the meaning of the adjective (red) with the meaning of the noun (apple). In two experiments, participants were asked to judge whether noun-adjective word-pairs were semantically related (e.g., red apple) or not (e.g., lucky milk) while online transcranial magnetic stimulation (TMS) was administered over the right cerebellum or over a control site (vertex in Experiment 1 and visual cortex in Experiment 2). Cerebellar TMS caused a decrease in participants' accuracy for related word-pairs while accuracy for unrelated stimuli was not affected. A third experiment using a control task where subjects compared pairs of random letters showed no effect of TMS. Taken together these results indicate that the right cerebellum is involved specifically in the processing of semantically related stimuli. These results are consistent with theories that proposed the existence of a unified cerebellar function within motor and non-motor domains, as well with recent perspectives about cerebellar involvement in semantic memory and predictive cognition.

Adipocyte lipolysis drives acute stress-induced insulin resistance.

Stress hyperglycemia and insulin resistance are evolutionarily conserved metabolic adaptations to severe injury including major trauma, burns, or hemorrhagic shock (HS). In response to injury, the neuroendocrine system increases secretion of counterregulatory hormones that promote rapid mobilization of nutrient stores, impair insulin action, and ultimately cause hyperglycemia, a condition known to impair recovery from injury in the clinical setting. We investigated the contributions of adipocyte lipolysis to the metabolic response to acute stress. Both surgical injury with HS and counterregulatory hormone (epinephrine) infusion profoundly stimulated adipocyte lipolysis and simultaneously triggered insulin resistance and hyperglycemia. When lipolysis was inhibited, the stress-induced insulin resistance and hyperglycemia were largely abolished demonstrating an essential requirement for adipocyte lipolysis in promoting stress-induced insulin resistance. Interestingly, circulating non-esterified fatty acid levels did not increase with lipolysis or correlate with insulin resistance during acute stress. Instead, we show that impaired insulin sensitivity correlated with circulating levels of the adipokine resistin in a lipolysis-dependent manner. Our findings demonstrate the central importance of adipocyte lipolysis in the metabolic response to injury. This insight suggests new approaches to prevent insulin resistance and stress hyperglycemia in trauma and surgery patients and thereby improve outcomes.

Immediate and transgenerational effects of thymol supplementation, inactivated Salmonella and chronic heat stress on representative immune variables of Japanese quail.

Environmental challenges are integrated in the inmunoneuroendocrine interplay, impacting the immune system of the challenged individuals, and potentially implying transgenerational effects on their offspring. This study addressed whether dietary supplementation with thymol can modulate the immune response of adult Japanese quail when simultaneously exposed to an inoculum of inactivated Salmonella Enteritidis and a chronic heat stress (CHS). We also evaluated whether the experienced situations by adults can affect the immune response of their undisturbed offspring. In the parental generation, supplemented quail exposed to CHS had a higher inflammatory response and similar values of the heterophil/lymphocyte (H/L) ratio than those that were not supplemented. In their offspring, those chicks whose parents were exposed to CHS showed higher inflammatory response and lower antibody production. Regarding the H/L ratio, chicks whose parents were supplemented showed lower H/L ratio values. Dietary supplementation with thymol partially and positively modulated the inflammatory response and avoided H/L ratio alteration in the parental generation exposed to high environmental temperatures, suggesting these adults were better at dealing with the challenge. The lower H/L ratio values in the offspring suggests that chicks are more capable to deal with potential stressful situations associated with conventional breeding conditions.

The role of allyl ammonium salts in palladium-catalyzed cascade reactions towards the synthesis of spiro-fused heterocycles.

There is a continuous need for designing new and improved synthetic methods aiming at minimizing reaction steps while increasing molecular complexity. In this respect, catalytic, one-pot cascade methodologies constitute an ideal tool for the construction of complex molecules with high chemo-, regio-, and stereoselectivity. Herein, we describe two general and efficient cascade procedures for the synthesis of spiro-fused heterocylces. This transformation combines selective nucleophilic substitution (S2'), palladium-catalyzed Heck and C-H activation reactions in a cascade manner. The use of allylic ammonium salts and specific Pd catalysts are key to the success of the transformations. The synthetic utility of these methodologies is showcased by the preparation of 48 spiro-fused dihydrobenzofuranes and indolines including a variety of fluorinated derivatives.
