Understanding charged vesicle suspensions as Wigner glasses: dynamical aspects.

Suspensions of charged vesicles in water with added salt are widespread in nature and industrial production. Here we investigate, via Brownian dynamics simulations, a model that grasps the key features of these systems, with bidisperse colloidal beads interacting via a hard-core and an electrostatic double layer potential. Our goal is to focus on a set of interaction parameters that is not generic but measured in recent experiments, and relevant for a class of consumer products, such as liquid fabric softeners. On increasing the volume fraction in a range relevant to real formulation, we show that the dynamics become progressively slower and heterogeneous, displaying the typical signatures of an approaching glass transition. On lowering the salt concentration, which corresponds to increasing the strength and range of the electrostatic repulsion, the emergence of glassy dynamics becomes significantly steeper, and, remarkably, occurs at volume fractions well below the hard-sphere glass transition. The volume fraction dependence of the structural relaxation time at different salt concentration is well described by a functional law inspired by a recently proposed model [Proc. Nat. Acad. Sci., 112, 13762 (2015)]. According to our results, the investigated system may be thought of as a Wigner glass, i.e. a low-density glassy state stabilized by long-range repulsive interactions. Overall, our study suggests that glassy dynamics plays an important role in controlling the stability of these suspensions.

Comparative transcriptome analysis reveals the regulatory effects of acetylcholine on salt tolerance of Nicotiana benthamiana.

Salinity is a major cause of crop losses worldwide. Acetylcholine (ACh) can ameliorate the adverse effects of abiotic stresses on plant growth, including salinity stress; however, the underlying molecular mechanisms of this process are unclear. Here, seedlings of Nicotiana benthamiana grown under normal conditions or exposed to 150 mmol L NaCl salinity stress were then treated with a root application of 10 μM ACh. Exogenous ACh application resulted in the downregulation of the activity of the antioxidant enzymes, ascorbate peroxidase, and catalase. ACh-treated plants had lower levels of reactive oxygen species, including the superoxide anion radical and hydrogen peroxide. Transcriptome analysis indicated that ACh treatment under salt stress promoted the differential expression of 658 genes in leaves of N. benthamiana (527 were upregulated and 131 were downregulated). Gene ontology enrichment and Kyoto Encyclopedia of Genes and Genomes pathway analyses revealed that exogenous ACh application was associated with a substantial increase in the transcripts of genes related to cell wall peroxidases, xyloglucan endotransglucosylases or hydrolases, and expansins, indicating that ACh activates cell wall biosynthesis in salt-stressed plants. ACh also enhanced the expression of genes associated with the auxin, gibberellin, brassinosteroid, and salicylic acid signalling pathways, indicating that ACh induces the activation of these pathways under salt stress. Collectively, these findings indicate that ACh-induced salt tolerance in N. benthamiana seedlings is mediated by the inhibition of antioxidant enzymes, activation of cell wall biosynthesis, and hormone signalling pathways. Stress-induced genes involved in osmotic regulation and oxidation resistance were induced by ACh under salt stress. The genes whose transcript levels were elevated by ACh treatment in salt-stressed N. benthamiana could be used as molecular markers of the physiological status of plants under salt stress.

Bile salt-dependent lipase promotes the barrier integrity of Caco-2 cells by activating Wnt/β-catenin signaling via LRP6 receptor.

Bile salt-dependent lipase (BSDL) within intestinal lumen can be endocytosed by enterocytes and support the intestinal barrier function. However, the epithelial-supporting effect of this protein has not been verified in a human cell line and neither the direct signaling pathway nor the function of endocytosis in this process has been clearly identified. We sought to investigate the signaling pathway and the membrane receptor through which BSDL might exert these effects using intestinal epithelial cells. Caco-2 cells were treated with recombinant BSDL, and the barrier function, cell proliferation, and activation of the Wnt signaling pathway were assessed. The effect of Wnt signaling activation induced by BSDL and BSDL endocytosis was investigated in LRP6-silenced and non-silenced cells. Moreover, caveolae- and clathrin-dependent endocytosis inhibitors were also applied respectively to analyze their effects on Wnt signaling activation induced by BSDL. BSDL treatment increased the barrier function but not proliferation of Caco-2 cells. It also induced β-catenin nuclear translocation and activated Wnt target gene transcription. Moreover, in the Wnt pathway, BSDL increased the levels of non-phosphorylated-β-catenin (Ser33/37/Thr41) and phosphorylated-β-catenin (Ser552). Notably, the silencing of LRP6 expression impaired BSDL endocytosis and decreased BSDL-induced β-catenin nuclear translocation. The inhibition of BSDL endocytosis induced by caveolae-mediated endocytosis inhibitor was stronger than that by clathrin-mediated endocytosis inhibitor, and the Wnt signaling activation associated with its endocytosis was also most likely caveolae-dependent. Our findings suggested that LRP6, a canonical Wnt pathway co-receptor, can mediate BSDL endocytosis and then activate Wnt signaling in Caco-2 cells.

LITHORISK.COM: the novel version of a software for calculating and visualizing the risk of renal stone.

Estimation of state of saturation with stone-forming salt represents a reliable tool to assess the overall risk. The available methods are based on computer-assisted ab initio calculations. Our earlier method URSUS was subsequently substituted by Lithorisk®, a software including visualization of risk profiles. Unfortunately, Lithorisk does not adapt to new versions of Windows® and Macintosh® Apple, neither runs on smartphones or tablets. We propose a novel version of the software which can be directly used online on any device equipped by different operating systems. Upon online connection and after registration, the software is ready for unlimited accesses, in either Italian, English or French. After digiting input variables (urea and creatinine also included) in a fixed dashboard, state of saturation is promptly given. In addition to state of saturation (ß) with calcium oxalate, brushite and uric acid, ß struvite and cystine are available. Both input variables and ß results are graphically depicted as green or red horizontal bars to indicate recommended values. The software was implemented with equations allowing to omit sulphate and ammonium excretion for users with difficult access to these measurements. This simplified version, tested for ßCaOx and ßBsh on 100 urine samples showed close correlation with the full version. The software gives a list of total and free concentrations and soluble complex species distribution. Results can be printed or saved as PDF. So, we propose an easily accessible software to estimate state of saturation usable on any operating system and personal device.

Microsized Red Luminescent MgAlO:Mn Single-Crystal Phosphor Grown in Molten Salt for White LEDs.

A single-crystalline defect-less phosphor is desired for efficient luminescence of the therein doped optical activators. In this paper, microsized MgAlO:Mn single-crystal phosphors with bright red luminescence were grown in molten LiCl salt at 950 °C, for application in blue LED pumped white lighting. By comparing the phosphor formation from various Mg- and Al-bearing sources, both the template-formation and the dissolution-diffusion processes were evidenced to account for the formation of the microsized MgAlO:Mn crystallites. Using nano γ-AlO as the Al-bearing precursor, the uniform MgAlO:Mn microcrystallites with a {111} planes-exposed tetragonal bipyramid morphology were obtained. The photoluminescence property was studied at various temperatures, and Mg ↔ Al anti-site disorder induced luminescence broadening was discussed. The MnE → A transition in MgAlO shows a quite short luminescence wavelength peaking at 651 nm and ultrabroadband emission extending to 850 nm. The luminescence is relatively robust against thermal effect with relatively high thermal quenching temperature of 400 K and activation energy of 0.23 eV. Employing the red-emitting MgAlO:Mn crystallites, blue LED pumped white lighting prototypes were fabricated which simulate the solar-like spectrum and show neutral to warm white.
