Combination of ataxia telangiectasia and Rad3-related inhibition with ablative radiotherapy remodels the tumor microenvironment and enhances immunotherapy response in lung cancer.
We investigated the combined effects of ataxia telangiectasia and Rad3-related (ATR) inhibition, ablative radiotherapy, and immune checkpoint inhibitor (ICI) therapy against lung cancer. ATR inhibitor was administered combined with ablative radiotherapy to assess its radiosensitizing effect on lung cancer cells. Treatment response and survival were evaluated in vivo using A549 xenograft flank tumor and synchronous LLC lung and flank tumor mouse models. Mice received ablative radiotherapy (12 Gy/d for 2 d), ATR inhibitor, and ICI. The tumor microenvironment was assessed in irradiated flank and non-irradiated lung tumors. Programmed death-ligand 1 expression was upregulated after irradiation. ATR inhibition attenuated this upregulation. ATR inhibitor pretreatment decreased cell survival after irradiation by inhibiting DNA double-strand break repair, inducing mitotic cell death, and altering cell cycle progression. ATR inhibition enhanced radiation-induced damage-associated molecular patterns determined by high mobility group box 1 quantification and activated the cyclic GMP-AMP synthase-stimulator of interferon genes pathway. Combined ATR inhibition and ablative radiotherapy inhibited tumor growth and improved survival in mice. Adding ICI therapy further enhanced local antitumor effects, reducing the metastatic lung tumor burden and remodeling the tumor microenvironment through immunogenic cell death induction and enhanced immune cell infiltration. Triple therapy increased immune cell infiltration in distant non-irradiated lung tumors and stimulated the generation of protective T-cell immunity in splenocytes. Safety analysis showed minimal toxicity. ATR inhibition enhanced the efficacy of ablative radiotherapy and immunotherapy in lung cancer. Transcriptomics-guided optimization of vitamins to enhance erythromycin yield in saccharopolyspora erythraea.
Comparative transcriptomics uncovered distinct expression patterns of genes associated with cofactor and vitamin metabolism in the high-yielding mutant strain Saccharopolyspora erythraea HL3168 E3, as compared to the wild-type NRRL 2338. An in-depth analysis was conducted on the effects of nine vitamins, and it was determined that thiamine pyrophosphate (TPP), vitamin B2, vitamin B6, vitamin B9, vitamin B12, and hemin are key enhancers in erythromycin production in E3, increasing the erythromycin titer by 7.96-12.66%. Then, the Plackett-Burman design and the path of steepest ascent were applied to further optimize the vitamin combination for maximum production efficiency, enhancing the erythromycin titer in shake flasks by 39.2%. Otherwise, targeted metabolomics and metabolic flux analysis illuminated how vitamin supplementation modulates the central carbon metabolism with notable effects on the TCA cycle and methionine synthesis to augment the provision of energy and precursors essential for erythromycin synthesis. The relationship between menstrual cycle characteristics, premenstrual syndrome prevalence and blood phenylalanine level in women with PKU.
Phenylketonuria (PKU) results from a deficiency in phenylalanine hydroxylase, catalyzing the conversion of phenylalanine (Phe) to tyrosine. Premenstrual syndrome (PMS) consists of physical, behavioral, and emotional symptoms occurring during the last week of luteal phase. The aim of the study was to determine the incidence of PMS, and document menstrual cycle characteristics of PKU patients to reveal the relationship with blood Phe levels. The study was conducted on 74 patients with a mean age of 21.7 ± 5.4 years. The mean age at menarche was 12.7 ± 1.3 years and 82.4 % had regular menstrual cycles. The periods of most patients (47.2 %) lasted 4-5 days. Menstrual cycles of 21-28 days were reported by 73 %, less than 21 days by 8 %, and more than 28 days by 19 % of women. Menorrhagia and dysmenorrhea was observed in 6.7 % and 71.6 % respectively. Adherence to diet was lost in 7 patients during the menstrual period. No significant relationship was found between Phe levels and PMS symptoms ( > 0.05). According to PMSS subscales, 52.7 % of patients with PKU had depressive feelings, 16.2 % anxiety, 55.4 % fatigue, 52.7 % irritability, 28.3 % depressive thoughts, 39.1 % pain, 59.4 % changes in appetite, 28.3 % changes in sleeping habits and 43.2 % had swelling. The findings of the study revealed that PMS prevalence was 39.1 % among PKU women. A new methodology for evaluating the neighbor discovery time in schedule-based asynchronous duty-cycling wireless sensor networks.
Duty cycling is a fundamental mechanism for battery-operated wireless networks, such as wireless sensor networks. Due to its importance, it is an integral part of several Medium Access Protocols and related wireless technologies. In Schedule-based Asynchronous Duty Cycle, nodes activate and deactivate their radio interfaces according to a pre-designed schedule of slots, which guarantees overlapping uptime between two neighbors, independent of the offset between their internal clocks, making communication between them possible. This paper presents a new methodology for evaluating the Neighbor Discovery Time (NDT) of Schedule-based Asynchronous Duty Cycle. Differently from previous methodologies, it accounts for the possibility of the slots in the schedules of the two neighbors not being perfectly border-aligned - an unrealistic assumption in practice. By means of simulation, we show that not taking this under consideration can lead to an overestimate of the NDT by a factor of 2 depending on the particular scenario, thus justifying the importance of our work.•We propose a new subslot-based methodology for computing the NDT of a wakeup schedule used for asynchronous duty cycling.•It replaces the traditional slot-based methodology, by dividing slots into subslots, allowing for the analysis of non-integer clock offsets between nodes, and further allowing mathematical models to consider the more realistic continuous-time case.•Our validation data shows that the slot-based methodology may overestimate NDT by a factor of up to 2, making the proposed subslot-based methodology much more precise.Diego Passos, Beatriz Trabbold, Ricardo C Carrano, Cledson de Sousa

Development of diffuse cutaneous squamous cell carcinoma in situ after chemotherapy for ovarian carcinoma and remission with PD1-inhibitor.
This is a 74-year-old female, initially presenting with malignant pleural effusion, and evaluation revealed programmed cell death ligand 1 (PDL1)-positive stage IV high grade serous ovarian cancer. Following initiation of standard chemotherapy agents, carboplatin and paclitaxel, the patient developed a diffuse, itchy rash over her abdomen, back, and bilateral upper and lower extremities. Biopsy of the rash revealed a diffuse non-resectable cutaneous squamous cell skin carcinoma (cSCC) in situ. Induction Chemotherapy for Sinonasal Tumors in Patients Presenting with Brain Invasion and/or Neurological Deficits.
The aim of this study was to investigate the safety of induction chemotherapy (IC) for patients with sinonasal malignancies with brain invasion or a neurological deficit. We conducted a retrospective analysis of patients who underwent IC for sinonasal malignancies with intracranial invasion or a neurological deficit at a single tertiary cancer center from 1992 to 2020. In total, 460 patients with sinonasal malignancies were included in the study. Of the patients reviewed, 341 underwent IC and within this group 40 had brain invasion (BI) and 31 had a neurological deficit (ND) at presentation. The most prevalent malignancy was sinonasal undifferentiated carcinoma (BI 40%, ND 41.9%), followed by esthesioneuroblastoma (BI 27.5%, ND 9.7%). All tumors were stage T4 with the majority lacking nodal metastases (BI N0: 72.5%, ND N0: 77.5%). All patients completed at least two cycles of IC. Partial or complete response to IC was seen in 80% of BI and 71% of ND patients. No patients had cessation of treatment due to neurologic decline and none required urgent surgery. Five patients (12.5%) with BI and 2 (6.5%) with ND had interruption of IC for reasons other than neurological decline. In patients with ND, IC led to improvement of 54.5% NDs. In patients with sinonasal malignancies with BI or ND who underwent IC, no patients had cessation of treatment due to neurologic decline. In contrast, most patients had improvement of neurologic symptoms with IC. Deciphering craniopharyngioma subtypes: Single-cell analysis of tumor microenvironment and immune networks.
Craniopharyngiomas, including adamantinomatous (ACP) and squamous papillary (PCP) types, are challenging to treat because of their proximity to crucial pituitary structures. This study aimed to characterize the cellular composition, tumor tissue diversity, and cell-cell interactions in ACPs and PCPs using single-cell RNA sequencing. Single-cell clustering revealed diverse cell types, further classified into developing epithelial, calcification, and immune response for ACP and developing epithelial, cell cycle, and immune response for PCP, based on gene expression patterns. Subclustering revealed the enrichment of classical M1 and M2 macrophages in ACP and PCP, respectively, with high expression of pro-inflammatory markers in classical M1 macrophages. The classical M1 and M2 macrophage ratio significantly correlated with the occurrence of diabetes insipidus and panhypopituitarism. Cell-cell interactions, particularly involving CD44-SPP, were identified between tumor cells. Eco-inspired synthesis of MgO-infused g-CN nanocomposites from tulsi seeds for advanced photocatalytic environmental remediation.
This study introduces a novel approach to synthesizing magnesium oxide (MgO) nanoparticles through the use of (tulsi seed) extract combined with the thermal polymerization of MgO-doped graphitic carbon nitride (MgCN) nanocomposites. The nanocomposites were prepared at varying MgO concentrations (0.5 mM, 1.0 mM, 1.5 mM, and 2.0 mM) to optimize their properties. Comprehensive characterization of the synthesized MgO nanoparticles and MgCN nanocomposites was conducted using advanced analytical techniques, including UV-Vis spectroscopy, X-ray diffraction (XRD), scanning electron microscopy with energy dispersive X-ray mapping (SEM-EDX), Fourier-transform infrared spectroscopy (FTIR), and thermogravimetric analysis (TGA). The MgCN nanocomposite with 1.5 mM MgO demonstrated a high surface area of 98.287 m g, as determined by Brunauer-Emmett-Teller (BET) analysis. X-ray photoelectron spectroscopy (XPS) confirmed the presence of carbon and nitrogen elements, validating the integration of MgO into the nanocomposite matrix. High-resolution transmission electron microscopy (HRTEM) images depicted planar, stacked, and wrinkled structures characteristic of a graphitic-like material. Consistent with a Z-scheme heterojunction, the MgCN (1.5 mM) sample exhibited an enhanced morphology, increased surface area, improved visible light absorption, and reduced band gap. This particular nanocomposite displayed remarkable adsorption and photocatalytic degradation capabilities, achieving up to 98% removal of methylene blue and 54% removal of tetracycline antibiotics. Furthermore, it showed significant antibacterial activity against . Notably, the MgCN (1.5 mM) nanocomposite maintained its performance over four cycles, underscoring its potential for sustained application in wastewater treatment and the elimination of organic contaminants. The scavenging activity of the nanocomposites was also explored, revealing additional environmental benefits.
