Oxidation of cathepsin S by major chemicals of cigarette smoke.

Lung cysteine cathepsin S (CatS) that is a potent elastase plays a deleterious role in alveolar remodeling during smoke-induced emphysema. Despite the presence of a reactive nucleophilic cysteine (Cys25) within its active site, most of its elastinolytic activity is preserved after exposure to cigarette smoke extract (CSE), a major source of sulfhydryl oxidants. This result led us to decipher CatS resistance to major and representative CSE oxidants: hydrogen peroxide, formaldehyde, acrolein and peroxynitrite. CatS was inactivated by hydrogen peroxide, peroxynitrite and acrolein in a time- and dose-dependent manner, while formaldehyde was a weaker oxidant. Hydrogen peroxide, but not CSE, formaldehyde, and peroxynitrite impaired the autocatalytic maturation of pro-CatS, whereas acrolein prevented the formation of mature CatS without hindering the initial step of the two-step autocatalytic process. Far-UV CD spectra analysis supported that oxidation by CSE and hydrogen peroxide did not led to a structural alteration of CatS, despite a notable increase of protein carbonylation, a major hallmark of oxidative damage. Evaluation of the oxidation status of Cys25 by specific biotinylated redox sensing probes suggested the formation of sulfenic acid followed by a slower conversion to sulfinic acid after incubation with hydrogen peroxide. Addition of reducing reagents (dithiothreitol, glutathione and N-acetyl cysteine) led to a partial recovery of CatS activity following incubation with CSE, hydrogen peroxide and peroxynitrite. Current results provide some mechanistic evidence of CatS stability and activity in the presence of CSE, supporting its harmful contribution to the pathophysiology of emphysema.

Characterizations of Gene Alterations in Melanoma Patients from Chinese Population.

Melanoma is a human skin malignant tumor with high invasion and poor prognosis. The limited understanding of genomic alterations in melanomas in China impedes the diagnosis and therapeutic strategy selection. We conducted comprehensive genomic profiling of melanomas from 39 primary and metastatic formalin-fixed paraffin-embedded (FFPE) samples from 27 patients in China based on an NGS panel of 223 genes. No significant difference in gene alterations was found between primary and metastasis melanomas. The status of germline mutation, CNV, and somatic mutation in our cohort was quite different from that reported in Western populations. We further delineated the mutation patterns of 4 molecular subgroups (BRAF, RAS, NF1, and Triple-WT) of melanoma in our cohort. BRAF mutations were more frequently identified in melanomas without chromic sun-induced damage (non-CSD), while RAS mutations were more likely observed in acral melanomas. NF1 and Triple-WT subgroups were unbiased between melanomas arising in non-CSD and acral skin. BRAF, RAS, and NF1 mutations were significantly associated with lymph node metastasis or presence of ulceration, implying that these cancer driver genes were independent prognostic factors. In summary, our results suggest that mutational profiles of malignant melanomas in China are significantly different from Western countries, and both gene mutation and amplification play an important role in the development and progression of melanomas.

Immunohistochemical evidence of canine morbillivirus (canine distemper) infection in coatis (Nasua nasua) from Southern Brazil.

The pathological and immunohistochemical (IHC) findings associated with infection due to canine morbilivírus (canine distemper virus, CDV) are described in coatis (Nasua nasua). Tissue fragments of coatis (n = 13) that died at the Bela Vista Sanctuary, Paraná, Southern Brazil, were routinely processed for histopathology to identify the main histopathologic patterns as compared to that of the domestic dog. Selected formalin-fixed paraffin-embedded (FFPE) tissue fragments of the lungs, liver, urinary bladder and small intestine were used in IHC assays designed to identify the antigens of CDV, canine adenovirus (CAdV-1 and CAdV-2) and canine parvovirus type 2 (CPV-2). The main histopathologic patterns identified were interstitial pneumonia (n = 9), interstitial nephritis (n = 6), atrophic enteritis (n = 4) and ballooning degeneration of the uroepithelium (n = 3). Positive immunolabelling for intralesional antigens of CDV was identified in the lung with interstitial pneumonia (n = 3), in the intestine (n = 2) and in the degenerated epithelium of the urinary bladder (n = 2). Antigens of CPV-2, CAdV-1 and CAdV-2 were not identified in any FFPE tissue sections evaluated. These findings indicate that these wild carnivores were infected by a viral disease pathogen common to the domestic dog and develop similar histopathologic findings. Collectively, these findings suggest that these coatis were infected by CDV and can serve as a potential host for this infectious disease pathogen.

Association between indoor formaldehyde exposure and asthma: A systematic review and meta-analysis of observational studies.

About 339 million people worldwide are suffering from asthma. We aimed to investigate whether exposure to formaldehyde (FA) is associated with asthma, which could provide clues for preventive and mitigation actions. This article provides a systematic review and meta-analysis of observational studies to assess the association between indoor FA exposure and the risk of asthma in children and adults. An electronic search of PubMed, Embase, and Web of Science was performed to collect all relevant studies published before January 1, 2020, and a total of 13 papers were included in this meta-analysis. A random-effect model was conducted to calculate the pooled odds ratio (OR) between FA exposure and asthma. We found that each 10 µg/m increase in FA exposure was significantly associated with a 10% increase in the risk of asthma in children (OR=1.10, 95% confidence interval=1.00-1.21). We sorted the FA concentrations reported in the selected articles and categorized exposure variables into low (FA≤22.5µg/m ) and high exposure (FA >22.5 µg/m ) according to the median concentration of FA. In the high exposure adult group, FA exposure may also be associated with an increased risk of asthma (OR=1.81, 95% CI=1.18-2.78).

Prognostic impact of PD-L1 in oropharyngeal cancer after primary curative radiotherapy and relation to HPV and tobacco smoking.

Incidence of oropharyngeal squamous cell carcinoma (OPSCC) is rising rapidly in many western countries due to (HPV) and tobacco smoking, with a considerable overlap. Immunotherapy directed at the PD1/PD-L1 axis have shown promise in head and neck cancer and other cancer types. PD-L1 expression may indicate a poorer prognosis, and at the same time indicate a possible benefit of anti-PD-L1 immunotherapeutic agents. The primary aim of this study was to establish the prognostic effect of PD-L1 expression after primary curative radiotherapy alone. A cohort of 303 OPSCC patients treated with primary, curative intended radiotherapy was established. PD-L1 expression was evaluated by immunohistochemistry on formalin fixed, paraffin embedded tissue sections. PD-L1 positivity was defined as a Combined Positive Score (CPS) ≥1, indicating staining of either tumor cells, lymphocytes or macrophages. Median follow-up was 5.3 years. With 199 deaths, there was no difference in overall survival between patients with PD-L1+ and PD-L1- tumors (adjusted hazard ratio [aHR] and 95% confidence interval [CI]: 1.0 [0.71-1.4]). Also, locoregional failure was similar between the two groups (aHR 1.1 [CI: 0.68 - 1.7]). Tumors were PD-L1+ in 76% of cases, significantly more among HPV p16+ tumors (82% vs. 70%,  = .01). Interestingly, higher prevalence of PD-L1+ expression was seen in HPV p16+ patients with <10 pack-years of tobacco-smoking (93%) compared to HPV p16+ smokers (76%) or HPV p16-negative patients (70%) ( = .003). PD-L1 expression had no prognostic significance in OPSCC patients treated with primary radiotherapy alone. A substantial proportion of OPSCC tumors show PD-L1 overexpression, especially in HPV p16+ tumors in patients with little or no smoking history.

A TP-FRET-based Fluorescent Sensor for Ratiometric Detection of Formaldehyde in Real Food Samples, Living Cells, Tissues and Zebrafish.

Formaldehyde (FA, HCHO) is a highly reactive carbonyl species (hRCS), which is very harmful to humans and environments as a tissue fixative and preservative. Therefore, developing some highly sensitive, selective and rapid detection methods is significant for human health in food safety and environmental protection. Herein, a two-photon (TP) ratiometric sensor CmNp-CHO has been constructed by conjugating a TP donor (Π-push-pull-structure) with a FA off-on acceptor (functioned with hydrazides moiety) via a non-conjugated linker through fluorescence resonance energy transfer (FRET) mechanism. Such the scaffold affords CmNp-CHO a reliable and specific probe for detecting FA with two-well-resolved emission peaks separated by 124 nm. And it responds to FA rapidly with high selectivity and sensitivity during 1.0 minute, and a large ratio enhancement at I550/I426 with addition of 0-20μM FA, exhibiting~4-fold ratio increase and a fairly low LOD of 8.3±0.3 nM. Moreover, CmNp-CHO has been successfully employed for detecting FA in live cells, onion tissues and zebrafish, which exhibiting CmNp-CHO can be serve as a useful tool for investigating FA in real food application, and offering strong theoretical support and technical means for investigation of physiological and pathological function of FA.

Recovery of electro-mechanical properties inside self-healing composites through microencapsulation of carbon nanotubes.

We report the successful microencapsulation of multi-walled carbon nanotubes suspended in a 5-ethylidene-2-norbornene (5E2N) self-healing monomer, into poly melamine urea formaldehyde shells through in situ polymerization. The average size of the microcapsules, their size-distribution, shell wall structural integrity and thickness are  characterized by optical and scanning electron microscopy. The presence of carbon nanotubes (CNTs) inside the core liquid content, as well as their release after breaking is confirmed by microscopy and spectroscopy analyses. A small amount of CNTs inside the microcapsules is found to have no significant impact on the thermal stability of the system, as determined by thermogravimetric analysis and differential scanning calorimetry. Both the mechanical and the electrical properties of CNT-based self-healing materials can be restored  up to 80% when CNT/5E2N microcapsules are incorporated into polymer composites, thus making them highly suitable for applications in aerospace.
