Characterization of bromelain indicates a molar excess of inhibitor vs. enzyme molecules, a Jacalin-like lectin and Maillard reaction products.

The phytotherapeutic bromelain is a heterogeneous protein mixture, extracted from pineapple stem, with high proteolytic activity based on cysteine proteases. Its global protein chemical composition was analyzed qualitatively and quantitatively by SDS-PAGE and RP-HPLC. A SDS-PAGE method with elaborate sample pretreatment was developed, to cope with the bromelain's self-digestion properties and the hypothetical disulfide scrambling during electrophoresis. Both can produce misleading results, if not considered. RP-HPLC was applied for its high separation power for bromelain proteinaceous compounds. A peak identification and assignment to different protein classes in bromelain was done by enzyme kinetics and MS. The method was successfully applied for the quantitative determination of the molar ratio between inhibitor and enzyme and resulted to be approximately 3:2. Bromelain contains, from a molar point of view, inhibitor molecules as major component, which thus might be considered as a natural pharmaceutical excipient in Bromelain, because it protects the enzymes against autolysis. We described two methods to separate the inhibitor fraction from the enzyme fraction, RP-HPLC and size exclusion chromatography. A pineapple derived Jacalin-like-lectin, herein called 'Anlec', was identified and quantified by RP-HPLC-MS in bromelain and its content was determined to be 5%, related to all proteins in bromelain. Anlec binds specifically to mannose-containing glycans and is discussed in literature to possess anti-HIV medical potential. Bromelain could therefore be a possible and economic source for the production of Anlec. An isolation strategy of Anlec from bromelain, in high purity, is shown in this work. The presented RP-HPLC results are comprehensive in chemical information, and the method is expedient to provide appropriate bromelain protein isolations but also to accomplish quality control, covering all relevant protein components. It is furthermore shown, that proteins in bromelain may react with reducing sugars in a Maillard reaction to form glycated proteins. Maillard reaction products in bromelain are detected and characterized and could be responsible for the limited stability and storage times at room temperature of bromelain. Even the active center thiol group could be potentially glycated.

Microbiome and diversity indices during blood stem cells transplantation - new perspectives?

The human body is colonized by bacteria, fungi and viruses. Resident commensal bacteria are a fundamental line of resistance to colonization by exogenous microbes. They actively regulate the production of nutrients by the host through a negative feedback mechanism, in order to prevent the availability of nutrients for potential pathogens. While only a small fraction of these microorganism may be pathogenic, the relationship between host and commensal microbiome is now studied as a whole, impacting several aspects of the host biology. Some studies have made clear the progresses in examining the role of microbiome on transplants and graft versus host disease (GVHD) severity and its pathogenesis: the risk of complications from allogenic hematopoietic stem cells transplantation (HSCT) is greater with the highest mortality if a patient has a lower bacterial diversity in the gut prior to the transplantation process beginning. Microbiota-associated molecular patterns are directly recognized by pathogen recognition receptors. The development of molecular methods has greatly expanded our knowledge of the composition and function of the microbiome in health and disease, shortening the response times vs. microbiological culture tests. The gut flora can make the difference when it comes to allo-HSCT. The aim of the study was to monitor microbiome of 10 children during allo-HSCT.

Scaling Behaviors of a Polymer Ejected from a Cavity through a Small Pore.

Langevin dynamics simulations are performed to investigate ejection dynamics of spherically confined flexible polymers through a pore. By varying the chain length N and the initial volume fraction ϕ_{0} of the monomers, two scaling behaviors for the ejection velocity v on the monomer number m in the cavity are obtained: v∼m^{1.25}ϕ_{0}^{1.25}/N^{1.6} for large m and v∼m^{-1.4} as m is small. A robust scaling theory is developed by dividing the process into the confined and the nonconfined stages, and the dynamical equation is derived via the study of energy dissipation. After trimming the prior stage related to the escape of the head monomer across the pore, the evolution of m is shown to be well described by the scaling theory. The ejection time exhibits two proper scaling behaviors: N^{(2/3ν)+y_{1}}ϕ_{0}^{-(2/3ν)} and N^{2+y_{2}} under the large and small ϕ_{0} or N conditions, respectively, where y_{1}=1/3, y_{2}=1-ν, and ν is the Flory exponent.

Discovery of an Exceptionally Strong β-Decay Transition of ^{20}F and Implications for the Fate of Intermediate-Mass Stars.

A significant fraction of stars between 7 and 11 solar masses are thought to become supernovae, but the explosion mechanism is unclear. The answer depends critically on the rate of electron capture on ^{20}Ne in the degenerate oxygen-neon stellar core. However, because of the unknown strength of the transition between the ground states of ^{20}Ne and ^{20}F, it has not previously been possible to fully constrain the rate. By measuring the transition, we establish that its strength is exceptionally large and that it enhances the capture rate by several orders of magnitude. This has a decisive impact on the evolution of the core, increasing the likelihood that the star is (partially) disrupted by a thermonuclear explosion rather than collapsing to form a neutron star. Importantly, our measurement resolves the last remaining nuclear physics uncertainty in the final evolution of degenerate oxygen-neon stellar cores, allowing future studies to address the critical role of convection, which at present is poorly understood.

THz Generation from Relativistic Plasmas Driven by Near- to Far-Infrared Laser Pulses.

Terahertz pulse generation by ultraintense two-color laser fields ionizing gases with near- to far-infrared carrier wavelength is studied from particle-in-cell simulations. For a long pump wavelength (10.6  μm) promoting a large ratio of electron density over critical, photoionization is shown to catastrophically enhance the plasma wakefield, causing a net downshift in the optical spectrum and exciting THz fields with tens of GV/m amplitude in the laser direction. This emission is accompanied by coherent transition radiation (CTR) of comparable amplitude due to wakefield-driven electron acceleration. We analytically evaluate the fraction of CTR energy up to 30% of the total radiated emission including the particle self-field and numerically calibrate the efficiency of the matched blowout regime for electron densities varied over three orders of magnitude.

Biomarkers in patients with heart failure and central sleep apnoea: findings from the SERVE-HF trial.

The Treatment of Sleep-Disordered Breathing with Predominant Central Sleep Apnoea by Adaptive Servo Ventilation in Patients with Heart Failure trial investigated the effects of adaptive servo-ventilation (ASV) (vs. control) on outcomes of 1325 patients with heart failure and reduced ejection fraction (HFrEF) and central sleep apnoea (CSA). The primary outcome (a composite of all-cause death or unplanned HF hospitalization) did not differ between the two groups. However, all-cause and cardiovascular (CV) mortality were higher in the ASV group. Circulating biomarkers may help in better ascertain patients' risk, and this is the first study applying a large set of circulating biomarkers in patients with both HFrEF and CSA.
