Health benefits of and subspecies BB-12 in children.

While the literature indicates that probiotics are effective for the treatment or prevention of several conditions, data in children specifically focusing on individual probiotic strains are still limited. Therefore, we conducted a narrative review that summarizes data on the efficacy and safety of two common probiotic strains- GG (LGG) and subspecies BB-12 (BB-12)-when administered to children. Our search of the PubMed database identified 13 meta-analyses, three systematic reviews, and 15 randomized, controlled trials that assessed BB-12 and LGG, either alone or in combination, when administered to infants to improve growth or to children of any age to prevent or treat acute gastroenteritis (AGE), antibiotic- or healthcare-associated diarrhea (AAD and HAD, respectively), respiratory infections, otitis media, and functional gastrointestinal (GI) disorders including irritable bowel syndrome (IBS). Our review found evidence that LGG can prevent AAD and improve the symptoms of AGE, particularly among children in Europe. However, we found only moderate evidence regarding the benefits of LGG for treating respiratory infections and IBS in children and minimal evidence to support the use of BB-12. While no serious safety concerns were identified, LGG was associated with two non-serious safety concerns of wheezing and viral skin infections. In conclusion, we propose that LGG can be safely administered to toddlers and older children to manage the symptoms of AGE and prevent AAD. These findings should be considered for incorporation into clinical practice guidelines and should help to focus the efforts of the probiotic industry.

Genetic variants in the TORC2 gene promoter and their association with body measurement and carcass quality traits in Qinchuan cattle.

The TORC2 gene is responsible for nutrient metabolism, gluconeogenesis, myogenesis and adipogenesis through the PI3K-Akt, AMPK, glucagon and insulin resistance signaling pathways. Sequencing of PCR amplicons explored three novel SNPs at loci g.16534694G>A, g.16535011C>T, and g.16535044A>T in the promoter region of the TORC2 gene in the Qinchuan breed of cattle. Allelic and genotypic frequencies of these SNPs deviated from Hardy-Weinberg equilibrium (HWE) (P < 0.05). SNP1 genotype GG, SNP2 genotype CT and SNP3 genotype AT showed significantly (P <0.05) larger body measurement and improved carcass quality traits. Haplotype H1 (GCA) showed significantly (p<0.01) higher transcriptional activity (51.44%) followed by H4 (ATT) (34.13%) in bovine preadipocytes. The diplotypes HI-H3 (GG-CC-AT), H1-H2 (GG-CT-AT) and H3-H4 (GA-CT-TT) showed significant (P<0.01) associations with body measurement and improved carcass quality traits. Analysis of the relative mRNA expression level of the TORC2 gene in different tissues within two different age groups revealed a significant increase (P<0.01) in liver, small intestine, muscle and fat tissues with growth from calf stage to adult stage. We can conclude that variants mapped within TORC2 can be used in marker-assisted selection for carcass quality and body measurement traits in breed improvement programs of Qinchuan cattle.

The role of genotype/phenotype at apurinic/apyrimidinic endonuclease Rs1130409 in renal cell carcinoma.

The DNA repair capacity plays a critical role in maintaining the genomic stability and gatekeeping for individual cancer risk. In this study, we aim at evaluation the role of the AspGlu (rs1130409) variant at apurinic/apyrimidinic endonuclease (APE) gene in renal cell carcinoma (RCC) risk and the contribution of different genotypes to its transcriptional mRNA levels. In the case-control study, 92 RCC patients and 580 cancer-free patients matched by age and gender were recruited. The apurinic/APE genotyping work was conducted with typical restriction fragment length polymorphism methodology after polymerase chain reaction. At the meanwhile, thirty renal tissue samples with variant genotypes were examined for their apurinic/APE mRNA and protein expressions by real-time quantitative reverse transcription method and Western blotting. The results showed that compared with the wild-type TT genotype, the people with TG and GG genotypes of apurinic/APE AspGlu had 0.88- and 1.09-fold risk of RCC, respectively. We have also examined the in vivo transcriptional (RNA) and translational (protein) levels with renal tissues of various apurinic/APE AspGlu genotypes, revealing that the apurinic/APE mRNA and protein were of similar levels among people of TT, TG, or GG genotypes. There was no joint gene-environment effect of apurinic/APE AspGlu genotype and smoking habit on RCC risk. The evidence indicated that apurinic/APE AspGlu genotypic variants did not alter its mRNA and protein expression among RCC patients. The genotype of apurinic/APE AspGlu may not serve as a proper predictive marker for RCC risk in Taiwan.

Comparing Interfacial Trp, Interfacial His and pH Dependence for the Anchoring of Tilted Transmembrane Helical Peptides.

Charged and aromatic amino acid residues, being enriched toward the terminals of membrane-spanning helices in membrane proteins, help to stabilize particular transmembrane orientations. Among them, histidine is aromatic and can be positively charged at low pH. To enable investigations of the underlying protein-lipid interactions, we have examined the effects of single or pairs of interfacial histidine residues using the constructive low-dynamic GWALP23 (acetyl-GGALWLALALALALALALWLAGA-amide) peptide framework by incorporating individual or paired histidines at locations 2, 5, 19, or 22. Analysis of helix orientation by means of solid-state H NMR spectra of labeled alanine residues reveals marked differences with H compared to W. Nevertheless, the properties of membrane-spanning HWALP23 helices show little pH dependence and are similar to those having Gly, Arg, or Lys at positions 2 and 22. The presence of H5 or H19 influences the helix rotational preference but not the tilt magnitude. H5 affects the helical integrity, as residue 7 unwinds from the core helix; yet, once again, the helix orientation and dynamic properties show little sensitivity to pH. The overall results reveal that the detailed properties of transmembrane helices depend upon the precise locations of interfacial histidine residues.
