XPF-ERCC1 protects liver, kidney and blood homeostasis outside the canonical excision repair pathways.

Loss of the XPF-ERCC1 endonuclease causes a dramatic phenotype that results in progeroid features associated with liver, kidney and bone marrow dysfunction. As this nuclease is involved in multiple DNA repair transactions, it is plausible that this severe phenotype results from the simultaneous inactivation of both branches of nucleotide excision repair (GG- and TC-NER) and Fanconi anaemia (FA) inter-strand crosslink (ICL) repair. Here we use genetics in human cells and mice to investigate the interaction between the canonical NER and ICL repair pathways and, subsequently, how their joint inactivation phenotypically overlaps with XPF-ERCC1 deficiency. We find that cells lacking TC-NER are sensitive to crosslinking agents and that there is a genetic interaction between NER and FA in the repair of certain endogenous crosslinking agents. However, joint inactivation of GG-NER, TC-NER and FA crosslink repair cannot account for the hypersensitivity of XPF-deficient cells to classical crosslinking agents nor is it sufficient to explain the extreme phenotype of Ercc1-/- mice. These analyses indicate that XPF-ERCC1 has important functions outside of its central role in NER and FA crosslink repair which are required to prevent endogenous DNA damage. Failure to resolve such damage leads to loss of tissue homeostasis in mice and humans.

REPORTED METHANE EMISSIONS FROM ACTIVE OIL AND GAS WELLS IN PENNSYLVANIA, 2014-2018.

Oil/gas well integrity failures are a common but poorly constrained source of methane emissions to the atmosphere. As of 2014, Pennsylvania requires gas and oil well operators to report gas losses, both fugitive and process, from all active and unplugged abandoned gas and oil wells. We analyze 589,175 operator reports and find that lower-bound reported annual methane emissions averaged 22.1 Gg (-16.9, +19.5) between 2014 and 2018 from 62,483 wells, an average of only 47% of the statewide well inventory for those years. Extrapolating to the 2019 oil and gas well inventory yields well average emissions of 55.6 Gg CH4. These emissions are not currently included in the state's oil and gas emissions inventory. We also assess compliance in reporting among operators and note anomalies in reporting and apparent workarounds to reduce reported emissions. Suggestions for improving the accuracy and reliability in reporting and reducing emissions are offered.

The association between Interleukin-6 rs1800795/rs1800797 polymorphisms and risk of rotator cuff tear in a Chinese population.

Expression of proinflammatory cytokines, such as interleukin-6 (IL-6) and metalloproteases, are elevated in patients with rotator cuff tear (RCT). In order to investigate the role of IL-6 gene polymorphisms on RCT risk, we genotyped two SNPs on IL-6 gene (rs1800795 and rs1800797) in 138 RCT patients  and 137 healthy controls  using PCR and sanger sequencing. The IL-6 expression in shoulder joint synovial fluid was determined by using Elisa method. The constant score and visual analog scale (VAS) were used to evaluate the clinical outcome of two treatment (surgical vs. conservative) for RCT patients. For rs1800795, individuals with the GG genotype or G allele had significantly higher risk of RCT. Elevated risk of tear size was associated with the GG genotype of the rs1800795 polymorphism. The IL-6 rs1800797 polymorphism was also associated with an increased risk of RCT, especially among female, drinkers, and individuals with BMI<25 kg/m2. The elevated levels of IL-6 gene were observed among the mutant genotype of rs1800795/rs1800797 polymorphism. Surgical group is significantly better than conservative treatment from the perspective of constant score and VAS. Furthermore, CG genotype of rs1800795 polymorphism increased the constant score at six months in comparison with CC genotype. In conclusion, our study supports a role of IL-6 rs1800795/rs1800797 polymorphisms on increased RCT risk. The RCT patients withCG genotype of rs1800795 polymorphism have more obvious surgical treatment effects by influencing the IL-6 expression.

Associations between gene polymorphisms and dilated cardiomyopathy in a Chinese population.

The present study aimed to investigate the role of gene polymorphisms in dilated cardiomyopathy (DCM) susceptibility and prognosis in a Chinese population. A total of 343 DCM patients and 451 controls were enrolled and had their blood genotyped. Survival analysis was evaluated with Kaplan-Meier curves and Cox regression analysis. G carriers (AG/GG) and AG genotype of rs237028 had a higher DCM susceptibility as well as a worse DCM prognosis. Additionally, C carriers (CT/CC) of rs652921 and G carriers (TG/GG) of rs521845 had a higher DCM risk and CC homozygote of rs652921 had a worse DCM prognosis. These associations were still significant after adjustment for the Bonferroni correction. gene polymorphisms were associated with DCM susceptibility and prognosis in the Chinese population.

The polymorphisms of IL-6/STAT3 signaling pathway may contribute to cutaneous T-cell lymphomas susceptibility.

IL-6/STAT3 signaling pathway has been suggested to play a role in CTCL pathogenesis. Polymorphisms in STAT3 signaling pathway-related genes might be a risk factor for CTCL. However, the exact role of inherited gene polymorphisms of IL-6 and STAT3 in the pathogenesis of CTCL is still not fully understood. The aim was to examine whether IL-6 cytokine and polymorphisms of IL-6 and STAT3 gene are associated with CTCL susceptibility, stage of disease and pruritus intensity. We compared the IL-6 serum level and the frequency of selected single nucleotide polymorphisms of IL-6 and STAT3 in 106 CTCL and 198 control group using polymerase chain reaction with sequence-specific primers method and ELISA. We have found that serum IL-6 level in CTCL patients was significantly higher than in healthy controls (p < 0.05). We also demonstrated that two genotypes, CC of IL-6 and GG of STAT3, were overexpressed in CTCL patients compared to healthy controls, and that they increase the risk of malignancy development (OR = 1.8, p = 0.04 for IL-6 and OR 2.53, p = 0.0064 for STAT3). Moreover, the GG genotype of STAT3 polymorphism seems to be associated with lack of pruritus or mild pruritus in CTCL patients. Our results indicate that IL-6 is involved in pathogenesis of CTCL but not pruritus. Moreover, CC of IL-6 and GG genotype of STAT3 genes might be considered as the risk factor for development of CTCL.

Role of rs4072037 polymorphism in gastric cancer: a meta-analysis.

To further determine the association between mucin 1 () rs4072037 polymorphism and gastric cancer risk on the basis of previously published studies. PubMed and Embase were used to search all the available publications. The relative risk of the correlation was shown as odds ratio (OR) with 95% confidence interval (95% CI) under all the genetic comparisons. Subgroup analyses based on ethnicity, study design and HWE were also executed to detect effects of specific factors on the risk of gastric cancer. rs4072037 polymorphism was observed to reduce the risk of gastric cancer under the five genetic comparisons [(GG versus AA: OR (95% CI)=0.72 (0.61, 0.84); GG + GA versus AA: OR (95% CI)=0.82 (0.76, 0.88); GG versus AA + GA: OR (95% CI)=0.83 (0.71, 0.96); G versus A: OR (95% CI)=0.78 (0.72, 0.84); GA versus AA: OR (95% CI)=0.80 (0.74, 0.87)]. This decreased risk of gastric cancer was also detected in subgroup analyses based on ancestry (Asian and Caucasian), study design (population-based and hospital-based) and HWE (>0.05). rs4072037 polymorphism may have an important role in gastric cancer, and this protective effect may vary among different ethnic populations and control subjects.

[A Meta-analysis of Pin1 gene polymorphism at -842 loci and cancer susceptibility].

Using Meta-analysis to evaluate the association between Pin1 gene polymorphism at -842 loci and cancer susceptibility. Pin1, polymorphism, tumor, variant and cancer as key words were used to systematically search for the case-control research on the association between the -842G/C polymorphisms of Pin1 and cancer susceptibility through China National Knowledge Infrastructure (CNKI), Wanfang Data, Embase and PubMed. The time of literatures was up to April 2(nd), 2019. Heterogeneity test, combined risk of cancer with the -842 C allele of Pin1, publication bias test and sensitivity analysis were performed by using Stata 12.0 software. A total of 144 articles were retrieved. According to the inclusion criteria, a total of 11 articles were included (2 Chinese documents and 9 English documents). There were 5 667 cases and 6 120 controls in eligible articles. The heterozygous model showed that Pin1 (-842G/C) polymorphism was associated with cancer susceptibility, and the pooled (95) value was 0.78 (0.61, 0.99). Subgroup analysis by cancer type suggested that the Pin1 (-842G/C) polymorphism could significantly decrease the incidence of breast cancer and lung cancer under the heterozygous model (GC vs GG), dominant model (GC+CC vs GG) and allele model (C vs G). The pooled (95) values were 0.73 (0.58, 0.92), 0.71 (0.57, 0.89), and 0.73 (0.60, 0.89) in breast cancer and 0.64 (0.52, 0.78), 0.64 (0.53, 0.78), and 0.67 (0.55, 0.80) in lung cancer. The variant -842 C allele could significantly increase the risk of nasopharyngeal carcinoma under the homozygote model (CC vs GG) and recessive model (CC vs GG+GC). The pooled (95) values were 2.22 (1.03-4.75) and 2.47 (1.16-5.26). No significant association was observed in squamous cell carcinoma. This Meta-analysis demonstrated that Pin1gene polymorphism at -842 was associated with cancer susceptibility.

Original behavior of L. rhamnosus GG encapsulated in freeze-dried alginate-silica microparticles revealed under simulated gastrointestinal conditions.

The probiotic bacteria L. rhamnosus GG (LGG) were encapsulated into core-shell alginate-silica microbeads of about 500 μm through a double step synthesis involving micro-ionogel formation by electrospraying and silica coating by the sol-gel process. Formulating microparticles with sucrose as a cryoprotectant allowed maintaining bacterial viability and cultivability upon freeze-drying for weeks, as determined by plate counting. As much as 3.4 × 10 CFU g and 4.1 × 10 CFU g were observed to be cultivable in alginate and silica-coated beads after 3 weeks of freeze-drying, respectively. The viability of the released bacteria was evaluated in an in vitro batch SHIME model (a simulator of the human intestinal microbial ecosystem) by denaturing gradient gel electrophoresis (DGGE) and quantitative PCR (qPCR). It was revealed that microencapsulation efficiently protects LGG from the low gastric pH, especially in the case of silica-coated beads. Both encapsulation systems (alginate and alginate-silica) allowed for a better colonization of the colon compared with free LGG. Interestingly, although metabolically inactive in the upper digestive tract, LGG released from silica-coated beads boost their metabolism once they arrive in the colon, where they outcompete other members from the Lactobacillus community. In view of these results, we show that silica, usually used as an anti-caking agent in food powders, can play an active role in probiotics delivery and colon colonization.
